Staining of human tissues for multi-color confocal microscopy analysis

This proposition describes a technique that allowed us to successfully stain human tissues with fluorochrome labelled antibodies for confocal and two-photon microscopy. Tissues are cut using either a microtome or a vibratome, fixed in parapholmaldeide and permebilized. This techinique is relatively complex and does not work for all the antibodies tested, the know-how to optimize the staining is relevant. The method can be of interest for partners interested in multi-parameter morphological analysis of human tissues.

Interest / Relevance: Study in human tissues of the activation of different cells of interest simultaneously, their reciprocal position, their contacts. For immunology the techniques allows to study immunological synapses in human explanted tissues.rnrnThe laboratory in Toulouse developed competitive facilities of microscopy and flow-cytometry.
