Mouse model for multiple sclerosis mediated by cd8 t cells

we generated mice in which the model antigen influenza hemagglutinin (HA) is selectively expressed as a neo-self antigen in oligodendrocytes, referred as MOG-HA mice. In these mice upon adoptive transfer of preactivated HA-specific CD8 T cells led to inflammatory lesions in the CNS, These lesions, associating CD8 T cell infiltration with focal loss of oligodendrocytes, and subsequent demyelination

Interest / Relevance: The loss of oligodendrocytes, the CNS myelin-producing cells, is a key feature of MS lesions, the precise contribution of CD8 T cells to oligodendrocyte death and to CNS demyelination is unknown. Therefore our mouse model provides a unique tool to study the direct effect of CD8 T cells on oligodendrocytes in vivo, to understand MS pathophysiology and to help drug development in MS.
